Analyzing the role of activated fibroblasts in the activation of macrophages

Abstract

Tumors are complex masses composed by multiple cell types, such as cancer associated fibroblasts (CAFs) and macrophages, that interact with each other promoting the tumor progression. Snail1 is a transcription factor required for the activation of CAFs. We have generated an MMTV-PyMT mouse model with inducible depletion in Snail1 gene, mostly expressed in CAFs, and observed that mice with Snail1 gene deletion presented a greater survival. Moreover, those tumors displayed fewer M2 macrophages. In this thesis, we showed that Snail1 was not expressed in macrophages and its deletion did not alter the in vitro polarization of monocytes towards M1 or M2 phenotype. The incubation of macrophages with Snail1-expressing CAFs or with their conditioned medium caused lower cytotoxic and phagocytic capability of macrophages towards tumor cells than when incubated with Snail1 KO CAFs (inactive CAFs). Furthermore, the analysis of the gene expression of macrophages polarized with CAF WT- versus CAF KO-conditioned medium revealed that active CAFs stimulated a complex combination of genes normally induced in an M2 phenotype, downregulated in M1 and other genes specifically regulated by active fibroblasts and not altered during the two classical differentiations. The inhibition of factors specifically released by active CAFs, such as prostaglandin E2 and TGFβ, reduced the expression levels of RNAs related to this M2-like polarization. Lastly, M2-like macrophages enhanced the tumor growth by promoting angiogenesis and the generation of an immune-suppressive microenvironment, inactivating CD8+ T cells and promoting the activation of regulatory T cells.

Els tumors són masses complexes formades per diferents tipus cel·lulars, com els fibroblasts associats al càncer (CAFs) i els macròfags, que interaccionen entre ells promovent la progressió tumoral. L’Snail1 és un factor de transcripció necessari per l’activació dels CAFs. Vam generar un model de ratolí MMTV-PyMT amb depleció induïble del gen Snail1, gen majoritàriament expressat en CAFs, i vam observar que els ratolins amb deleció al gen de Snail1 presentaven millor supervivència. A més, aquells tumors tenien menys macròfags M2. En aquesta tesis, vam ensenyar que Snail1 no s’expressava en macròfags i que la seva deleció no afectava la polarització in vitro dels monòcits cap a un fenotip M1 o M2. La incubació de macròfags amb CAFs que expressen Snail1 o amb el seu medi condicionat va induir menys capacitat citotòxica i fagocítica cap a les cèl·lules tumorals que no aquells que havien estat incubats amb CAFs sense Snail1 (CAFs inactius). A més a més, l’anàlisi de l’expressió gènica dels macròfags polaritzats amb medi condicionat de CAFs amb o sense Snail1 va revelar que els CAFs actius estimulaven una combinació complexa de gens normalment induïts en el fenotip M2, regulats a la baixa en el fenotip M1 i altres gens específicament regulats per fibroblasts actius però que no estan modificats en les dues diferenciacions clàssiques. La inhibició dels factors específicament alliberats pels CAFs actius, com la prostaglandina E2 i el TGFβ, reduïa els nivells d’expressió dels ARN relacionats amb aquesta polarització del tipus M2. Per últim, els macròfags de tipus M2 afavorien el creixement del tumor promovent la formació de vasos sanguinis i la generació d’un microambient immunosupressor, inactivant les cèl·lules T CD8+ i promovent l’activació de les cèl·lules T reguladores.
